The Varian Cary 100 is a double beam, dual chopper, recording spectrophotometer controlled by a computer operating under Windows 2000, and running WinUV software. The instrument has the following features, using 1-cm cells.
· 0.28 M Czerny-Turner monochromator with a 1200 lines/mm grating, wavelength range from 190 to 900 nm, R928 photomultiplier, sources are tungsten halogen visible light and deuterium arc for ultra-violet light. · A working range past 3.5 absorbance units - Dilution of most sample is not normally required. · The thermostated, four-position cell holder is suitable for DNA thermal denaturation/ renaturation, enzyme kinetics, membrane and protein studies with the Cary 100 and Eclipse fluorescence spectrophotometer. The Peltier cell holder features electromagnetic stirring and electronic heat pumps, known as Peltiers, that improve the accuracy and precision of temperature control. Only the cell compartment will be thermostatted. No cooling or heating will be observed in the transfer lines. An aqueous solution of antifreeze fills the heat pump, if the pump begins to make an unusual noise turn it off and add more coolant. Cary WinUV software has a modular design. The software applications are separated into modules specific for each application (e.g. Scanning, Kinetics, RNA/DNA, Concentration, Simple Reads, Validation, etc.). There are two large Start and Stop buttons for each procedure. All data gathering begin with Setup to change default values or saved parameters followed by zero background measurements.
