Other Procedure: Cary 100 -- UV and Visible Spectrophotometer

 

I. Obtaining Absorption Data

(See Scan for details on setup to select wavelength and other paramaters.)

1. Insert blank cell, filed 2/3 full with the solvent..
2. Click on the Zero button.
3. Remove the blank sample and insert the sample to be analyzed.
4. Click on the Read button. The absorption data will appear in the report space.
5. For running multiple samples, load the next sample and click on Read again. The data will appear in list form on the screen.

II. Measuring Absorption over Time (Kinetics)

Starting the Kinetics Program and Choosing a Method
1. Double click on the Kinetics icon in the Cary WinUV window.
2. From the File Menu in the menu bar, pick Open Method.
3. In the window that appears, double click on the folder with your method.
4. Choose your method from the folder and click Open. The method contains parameters that have been preset for the experiment.

Choosing a Wavelength
1. Click on Setup in the menu bar.
2. Find the Wavelength section on the Cary Instrument Control tab and type in the desired wavelength. Do not change any of the other parameters.
3. Click on OK.

Obtaining Data
1. Place your blank sample in the machine and click the Zero button.
2. Remove the blank and press the Start button located above the graph. In the dialog box that appears, click OK.
3. A 2 minute countdown is activated. Mix the components of the sample. Place the sample in the spectrophotometer, and immediately record data by clicking on OK in the countdown box. This is important when measuring initial rates.
4. The run time is set for 10 minutes. To stop a run before the time is up, click the Stop button located above the graph. Click OK in the dialog box that appears.

Saving Data

1. Choose Save Data As from the File Menu.
2. Go to the Save as Type section at the bottom of the window, and choose Spreadsheet ASCII (x.CSV) from the dropdown menu.
3. Double click on your folder, name the data, and click Save.
4. To save the data to a disk, go to Save in at the top of the window and choose 3.5 Floppy (A:) or memory stick (D:) from the pulldown menu. Name the data and click Save.

Printing Data
1. To print something saved in your folder, choose Explorer from the File Menu. Navigate to your data by double clicking first on the (C:) drive icon, then on the Varian folder, the Cary WinUV folder, and the chemistry or share folder. Double click on your data file to open it.
2. In the window that opens, choose Print from the File Menu to print your data.
3. To print data saved to a disk, open the file on your disk and choose Print from the File Menu
